Establishment of a Protoplasts-Based Transient Expression System in Banana (Musa spp.)

The breeding of triploid banana cultivars with improved traits, such as yield and disease resistance, remains a major challenge for breeders. One reason is that the molecular study functional gene analysis in bananas fall behind due to difficulties its genetic manipulation. plant protoplast-based transient transformation has been documented widely used versatile convenient system many species. However, an efficient high-quality protoplast isolation still lacking bananas. Here, we established method by selecting proper source materials, optimizing conditions enzymatic hydrolysis PEG-mediated transfection. We found best materials protoplasts’ are young suckers, which give protoplasts ranging from 2.5 × 106 10.1 107 g?1 fresh weight after 5 6 h enzymolysis. sufficient most assays have protoplasts-based systems, protein subcellular localization interaction assays. Moreover, using expression protoplasts, validated Arabidopsis VESICLE SORTING RECEPTOR 1 (VSR1) self-interaction CNGC20 on cell membrane. results indicated this works well could be routinely characterization genes.
